Case: ΦX174 Lytic Gene E
Gene E encodes a potent lytic protein that cleaves bacterial cell walls, causing 100% cell lysis.
Cloning this gene in standard E. coli strains failed due to leaky expression and interaction with the host protein SlyD, leading to premature cell death.
Small colonies and escape colonies ( large colonies )

Solution: Engineering a Neutral Host
By knocking out slyD, we created a safe cloning host where Gene E could be stably maintained. When transferred into SlyD-expressing strains, the gene functioned as intended, enabling controlled bacterial lysis.
